8. Image stack settings
Collect the following set of images
Collect additional images above | below the selection: Designate the desired distance in microns.
Collect Images for:
- Mounted thickness: Acquires the image stack for the entire specified mounted thickness.
- Probe and top guard zone: Acquires the image stack for the specified disector height (probe) and top guard zone only; it will not acquire the bottom guard zone.
- Probe only: Acquires the image stack for the specified disector height (probe) ONLY; it will not acquire the top and bottom guard zones.
Enter Image Stack height information
Top guard zone / Probe height: Must be less than the mounted thickness when combined.
Mounted thickness: Must be less than the section cut thickness.
If you don't know the exact mounted thickness and wish to determine it manually, click Set. The Measure tissue thickness dialog is displayed.

Identify the top and bottom of the section:

Focus to the top of the tissue and click Set top of section.

Focus to the bottom of the tissue and click Set bottom of section.
The height of the tissue is displayed.

Accept the measurement by clicking Add measurement to list. This sets the height as your Mounted thickness.
You can click:

Delete measurement to start again

Revisit measurement to edit your settings

Distance Between Images
Step Size: Enter the step size (µm) of your image stack.
