1. Set up the subject
Subject Information
Type your name, the subject name, and any other information that you think will be helpful in the future.
Sampling Parameters
Use an existing sampling configuration? Choose yes or no to use a previously saved sampling configuration or not.
If yes, click the browse icon. In the Sampling Parameter Chooser window, highlight the parameter set you wish to use and click OK.
Enter Section Information:
- Enter the distance between [section] pairs.
Example
If the cut section thickness (block advance) is 5 µm and you are using a periodicity of 10, the distance between pairs is 50 µm. For help determining the most suitable distance between pairs, Determining Sampling Precision
If you are using the Smooth Fractionator Method (Gundersen, 2002), keep note of the fraction you are using.
- Lookup Z position: The Lookup section is the second section in the pair.
- Reference Z position: The Reference section is the first section in the pair.
Example
- Cut thickness = 5 µm
- Adjacent pairs
- You are starting with the very first cut pair.
Then the Reference Z Position is "0" and the Lookup Z Position is "5."

Click the browse icon to load the appropriate Reference and Lookup images.
