Select the acquisition type for the 2D Virtual Tissue image.
Brightfield
This type may be more suitable to obtain a good contour, even if you plan on acquiring a multichannel fluorescent 3D Virtual Tissue.
Single channel
Requires a command sequence, typically to set up the light path by moving mirror cubes and filters.
Running a command sequence before each acquisition enables the persistence of camera settings.
Multichannel
To use this type, define command sequences with the Multichannel Acquire Setup button. See Acquire Setup: Automating tasks for the acquisition of multichannel images / Multichannel Acquires tab for details.
Use the same parameters for the 3D Virtual Tissue multichannel acquisition (step 9).
Multichannel settings do not persist.
Some lights require that you add a delay to the command sequence after setting the light level to give the light time to settle at the new level. Using such a command sequence at this step means that the delay only occurs once instead of occurring before each tile.
