Image Illumination Correction

Use Image Illumination Correction when working with images already acquired.

Be aware that:
Illumination correction is designed for fluorescent images -- applying this command to other types of images is likely to yield poor results.
This command is CPU-intensive.

  1. Open a fluorescent image/image stack.
  2.  Click Image>Image Illumination Correction. Neurolucida places a check mark next to the command and displays the Illumination Correction dialog box .
    • Type: Start with Gentle.
    • Light Removal: Start with 50%.
    • Target Channel: Apply to all channels or to an individual R,G,B channel.
  3. Click OK.
  4. Examine the effects of illumination correction. If you are not satisfied with the results:
    1. Close the image/image stack.
    2. Open the image/image stack again.
    3. Click Image>Image Illumination Correction again to select different settings.

How it works

In the absence of a background image, Neurolucida uses the image to estimate uneven illumination for background correction.
