Create new lens from image

Procedure

  1. Load an image file with embedded scale factor and aspect ratio.
  2. You may be prompted to determine the correction factor that is required for the next step.
  3. Go to Tools>Create New Lens to open the Create New Lens window.
  4. Modify the settings as needed and click OK.

    5. ClosedName
    • Choose a name that permits unambiguous identification of the lens.
    ClosedComments

    Add comments to help you identify the lens and its conditions of use. These comments are inserted into the lens calibration file (‘stereo.len’) and do not appear elsewhere.

    ClosedUnits

    Change to millimeters or nanometers for data tablet use or EM.

    ClosedScale factor

    Displays the factor detected in the file. If the value is incorrect, enter the correct value manually.

    ClosedAspect ratio

    Displays the ratio detected in the file. If the value is incorrect, enter the correct value manually.

    ClosedZ Correction Factor

    Used to calculate the true position of the focal plane as the microscope is moved up and down. The focal plane moves a distance that may differ from the movement of the microscope. The difference is due to the refraction of light between the cover slip and the immersion medium of the lens. Under normal circumstances the predefined values for air, oil, and water immersion lenses can be used. There are circumstances in which the predefined settings should not be used.     • Select User-Specified to enter your own correction factor.

    To enter a nonstandard correction factor use Other. The correction factor is usually the inverse of the index of refraction between the tissue and the lens immersion medium.
