4. Select auto-contour acquisition type
Select the acquisition type for the 2D slide scan.

Brightfield
This type may be more suitable to obtain a good contour, even if you plan on scanning a multichannel fluorescent 3D slide.
Single channel
Requires a command sequence, typically to set up the light path by moving mirror cubes and filters.
Running a command sequence before each acquisition enables the persistence of camera settings.
- Before acquire—Requires a command sequence even if the command sequence is "empty" (i.e., it won't lead to a change of state for any device). If there is no existing command sequence, use the Edit Command Sequences button (top right corner of the dialog) to create it.
- After acquire—Optional
Multichannel
To use this type, define command sequences with the Multichannel Acquire Setup button. See Automating the multichannel acquisition process for details.
Use the same parameters for the 3D multichannel slide scan (step 9).
Multichannel settings do not persist.
Command sequence before auto-contour acquire
- Select a command sequence from the drop-down menu to maintain a light at a constant setting through the entire 2D slide (auto-contour) scan.
Some lights require that you add a delay to the command sequence after setting the light level to give the light time to settle at the new level. Using such a command sequence at this step means that the delay only occurs once instead of occurring before each tile.
