Acquire BG (for background correction)

Purpose

Use the Aquire BG button to capture a background image that can be used to correct for and mitigate uneven illumination in acquired images.

Before you start

Review information about acquiring a background image, according to the imaging technique you are using:

  • Instructions for Brightfield microscopy

  • Instructions for Fluorescence microscopy

Procedure

  1. Click the Live Image button in the Display section of the Acquire ribbon.
  2. Click the Acquire BG button in the Background Correction section of the Acquire ribbon.
  3. In the Acquire Background Image window that opens, select the most suitable options (described below) and click OK.

Type of background correction

Select the type of background correction based on the imaging technique you are using.

Multichannel options

  • Multichannel: See "Video Tool Panels: Multichannel Control"
  • Pause between channels: Available if the Multichannel option is selected. Select this option to generate alerts each time a channel has been acquired, so that you can make any changes or adjustments to the image.

Enable background correction

This option is "on" by default. It is useful to correct illumination in brightfield images and extremely valuable in slide scanning . See Capturing a background image for slide scanning (brightfield only).
